Effect of microRNA-195 on the expression of CCND1 in glioma U251 cell

Abstract:

Objective

To research the effect of microRNA-195 on the expression of CCND1 in glioma U251 cell.

Methods

MicroRNA-195 mimics, microRNA-195 inhibitors were transfected into human glioma U251 cell by lipofectamine 2000 transient transfection. The microRNA-195 mimics transfection group was M group, and the microRNA-195 inhibitors transfection group was group I(blank control group, K group). Real-time PCR was used to detect the content of microRNA-195 before and after transfection to ensure the transfection efficiency. The expression of CCND1 mRNA was detected by real-time PCR. The expression of CCND1 in glioma U251 cells was detected by western blotting.

Results

Real-time PCR results showed: the expression of CCND1 mRNA in group M was lower than that in group I (M:758.264<I:1398.04) ; western blotting: the expression of CCND1 was decreased in M group compared with I group(M:2029<I:22365.66).

Conclusion

MicroRNA-195 has an effect on the expression of glioma U251 cell line CCND1.
