Effect of resveratrol on the formation of intracranial aneurysms and telomere length

Abstract:

Objective

To investigate the effects of resveratrol on the formation of intracranial aneurysm (IA) and telomere length in rats.

Methods

Thirty-six SD rats were randomly divided into control group (n=12), IA group (n=12) and resveratrol group (n=12). Rats in IA group and resveratrol group were used to construct IA rat model. According to the dose of 40 mg/kg, resveratrol was dissolved in 2 mL saline and given intragastrically in the resveratrol group rats, and 2 mL saline was given to the rats in the IA group every day, and no additional operation was performed in the control group. After 12 weeks, the relative length of telomere and the relative expression of telomerase reverse transcriptase (TERT) and telomere binding protein-tripeptidase 1 (TPP1) were measured by real-time PCR, and the formation of IA was observed by scanning electron microscope after vascular casting.

Results

There was no significant difference in the formation rate of IA between resveratrol group and IA group (75.0% vs 66.7%, P>0.05), but the formation rate of stage Ⅲ aneurysm in resveratrol group was significantly lower than that in IA group (χ2=4.800, P=0.028). The telomere length and the expression of TERT and TPP1 in IA group and resveratrol group were significantly lower than those in control group(P<0.05). Compared with IA group,resveratrol group had longer telomere length and higher expression of TERT and TPP1 (P<0.05).

Conclusion

Resveratrol has no effect on the formation of IA, but can inhibit the shortening of telomere length and delay the progress of IA.
