Effect and mechanism of JAZF1 in glioblastoma invasion and migration

Abstract:

Objective

To investigate the expression of JAZF1 in glioblastoma (GBM) and its effect of invasion and migration in GBM and the related mechanism.

Methods

By digging through TCGA database in different grade gliomas JAZF1 transcription level differences and differences in survival curve, and then by the glioma samples collected in the division of the total protein imprinting experiments JAZF1 protein levels in glioma differences of different levels, through the Trans-well experiment and scratched cell research JAZF1 influence on glioma cell invasion and migration, through experiments protein imprinting JAZF1 on glioma cell invasion and migration ability of the influence of key molecules.

Results

(1) The mRNA and protein level of JAZF1 in grade Ⅳ glioma (GBM) are significantly higher than grade Ⅱ and grade Ⅲ glioma (P<0.05); (2) The median survival time in JAZF1 high expression group is 13.0 months, but 30.2 months in JAZF1 low expression group, this difference is significant (P<0.05); (3) When JAZF1 expression in U87M is silenced, the ability of invasion and migration is degressive significantly; (4) After JAZF1 expression was disturbed, p-Akt, MMP9 and MMP2 expression levels were significantly down-regulated, with statistically significant differences (P<0.05), while Akt expression was not significantly changed (P>0.05).

Conclusion

JAZF1 is high expression in GBM and shorten the median survival time significantly, and JAZF1 regulate the expression level of p-Akt and related downstream protein expression level, MMP9 and MMP2, to control the ability of invasion and migration in GBM.
