Abstract:

Objective

To study the expression of Homer1a in brain diffuse axonal injury in rat pallium and brainstem.

Methods

Ninety SD rats, with weight of(200±20) g, were randomized to normal and DAI group. The rat DAI models were produced in DAI group and the normal group was treated in the same way given to the DAI group except for "free rotating" attack. The rats’pallium and brainsteam were detected for Homer1a by immunoblotting and immunohistochemistry after injury at 10 min, 30 min, 1 h, 6 h, 12 h, 24 h, 48 h, 72 h respectively.

Results

The Homer1a was positive in the pallium after injury from 30 min to 72 h. The Homer1a was positive in the brainstem after injury from 10 min to 72 h. The expression of Homer1a in pallium was weaker than in brainstem at every time we detected. Homer1a was no expression in normal group.

Conclusion

The expression of Homer1a in pallium and in brainsteam was significant difference, indicates that the pathognomonic of DAI was brain stem inury. The Homer1a maybe involved in the brain protection after neuronal injury.

Key words: Homer protein, Immunohistochemistry, Immunoblotting, Diffuse axonal injury
